ISOLASI FLAVONOID FRAKSI ETILASETAT DAUN BERINGIN (Ficus benyamina L.)

Abstract


ABSTRACT

The isolation of flavonoid compound from ethyl acetate fraction of beringin leaves (Ficus benyamina L.) havebeen done. The extraction was prepared with maseration method using methanol solvent, and then followed byfractination with liquid-liquid extraction using water, ethyl acetate, and n-hexane. Separation method was conductedby column chromatography using eluent gradation with blend of n-hexane and ethyl acetate, and thencontinued by preparative thin layer chromatography until the isolate obtained. The purity of isolate was testedwith two dimensional thin layer chromatography and showed one spot which indicated that isolate was pure. Thecharacterization of isolate was investigated with ultraviolet-visible spectrophotometry and infrared spectrophotometry.Ultraviolet-visible spectrophotometry resulted maximum wavelength 272,80 nm (band II) and 343,20(band I) that included of flavon compound, and infrared spectrophotometry was proved bond of O-H (3405,67cm-1), C-H (2919,7 cm-1), C=O (1639,2 cm-1), C=C (1461,78 cm-1), C-O (1103,08 cm-1).

ABSTRAK

Telah dilakukan isolasi golongan senyawa flavonoid fraksi etil asetat daun beringin (Ficus benyamina L.) yangdiperoleh dari Cimahi Jawa Barat. Flavonoid merupakan salah satu metabolit sekunder yang mempunyai banyakaktivitas biologi diantaranya aktivitas anti mikroba. Ekstraksi dilakukan dengan cara maserasi menggunakanpelarut metanol, dilanjutkan dengan ekstraksi cair-cair dengan pelarut n-heksana, etil asetat, air.Fraksi etil asetat dipisahkan dengan kromatografi kolom menggunakan eluen campuran n-heksana dan etilasetatsecara gradien dihasilkan empat fraksi gabungan F-I, F-II,F-III, F-IV. Fraksi-II (41-54) dipisahkan lanjutdengan kromatografi lapis tipis preparatif, pengembang n-heksana: etil asetat (3:5) diperoleh isolat dengan fluoresensi biru. Data karakterisasi isolat dengan spektrofotometri UV-Vis memberikan panjang gelombangmaksimum 272,8 nm pada pita II dan 343,2 nm pada pita I yang merupakan golongan flavon dan dan dari hasilspektrofotometri infra merah menunjukkan adanya ikatan O-H, C-H, C=O dan C=C.
