Abstract


ABSTRACTPiper betle L. (betel leaves) and red betel (Piper crocatum Ruiz & Pav) are medicinal plant that are often usedby the Indonesian people to treat various diseases, including cancer. Scientific evidence to strengthen then useas anticancer is still necessary. This study was conducted to determine the effect of green and red betel leafinfusion against HeLa cervical cancer cell viability, as well as its potential to inhibit the expression of Bcl2 protein qualitatively. Infusionof betel leaves were dried over a water bath to obtain a dry extract. Effect of extracts on theviability of HeLa cells determined by the cytotoxic test with MTT assay method to obtain IC50 values . Effect theactivity against Bcl2 protein was tested byimmunocyitochemical test. Cytotoxic test infution of green betel leavesgave IC50 value of 49.14 ug/ml, whereas the red betel showed no cytotoxic effects. The immunocytochemical testshowed a reduction in Bcl-2 protein expression.The results of this study showed that green betel leaves affect HeLacell viability by inducing cell death through inhibition of anti-apoptotic Bcl2 protein expression. Thus green betelleaves are potential to be developed as a chemopreventive agent.Keywords: Piper betle, Piper crocatum, cytotoxic, HeLa cells

 

ABSTRAKSirih hijau(Piper betle L.) dan sirih merah (Piper crocatum Ruiz & Pav) merupakan tanaman yang seringdimanfaatkan oleh masyarakat Indonesia untuk mengobati berbagai macam penyakit, termasuk kanker. Buktiilmiah untuk memperkuat dasar penggunaannya sebagai antikanker masih sangat diperlukan. Penelitian inidilakukan untuk mengetahui efek infusa daun sirih hijau dan sirih merah terhadap viabilitas sel kanker serviksHeLa, serta potensinya dalam menghambat ekspresi protein antiapoptosis Bcl2 secara immunositokimia.Infusa sirih dikeringkan di atas penangas air hingga diperoleh ekstrak kering. Pengaruh ekstrak terhadapviabilitas sel HeLa ditetapkan melaluiuji sitotoksik dengan metode MTT assay hingga diperoleh nilai IC50.Pengaruh ekstrak aktif terhadap protein Bcl2 dilakukan dengan pengecatan immunositokimia. Uji sitotoksikinfusa sirih hijau memberikan nilai IC50 49.14 μg/ml, sedangkan sirih merah tidak menunjukkan efek sitotoksik.Hasil pengecatan immunositokima secara kualitatif memperlihatkan pengurangan ekspresi protein Bcl2.Hasil penelitian ini menunjukkan bahwa sirih hijau mempengaruhi viabilitas sel HeLa dengan menginduksikematian sel melalui penghambatan ekspresi protein antiapoptosis Bcl2. Dengan demikian sirih hijau cukuppotensial dikembangkan sebagai agen kemopreventif untuk kanker serviks.
