ABSTRACTTemulawak (Curcuma xanthorriza) and Sambiloto (Andrographis paniculata) are known as Indonesian medicinalplant that has many properties and used for health for generations, one of them as an immunostimulant orendurance enhancer. The research objective was to determine the efficacy of herbal formulas development oftemulawak and sambiloto in improving endurance, i.e.by increasing the number of Th CD4+ cells. The study wasconducted using experimental animals ddY strain male mice (deutch democratic Yokohama) induced with 0.1 mli.p. BCG vaccine on the first day and the seventh, the extract was administered for 10 days. At the last days (H-10)levels of Th CD4+ was measured using flowcytometer. Experimental animals were grouped into 7 groups, eachgroup consists of 5 mices. The first and second groups were given extracts of sambiloto, temulawakorally at a doseof 3 mg/20 g bw. The third and fourth groups were administered with formula extracts of javanese turmeric andsambiloto with a dose of 6 and 30 g/20 g bw. While group of five, six, and seven are the positive control group,negative and carrier. Results showed that the extract of javanese turmeric and sambiloto formula increasedthe number of Th CD4+ cells compared to the carrier control (p<0.05). Population of Th CD4+ cells of sambilotoextract group (4.19x103/ml) was the highest, compared with the other groups (II. 2,34x103/ml; III. 2,13x103/ml;IV. 2,70x103/ml), positive (2.56x103/ml), and carrier (3.21x103/ml). It can be concluded that the single extractor formula extract of temulawak and sambilotowere able to increase the number of Th CD4+ cells compared withcarrier control (p <0,05).

Abstrak

(Curcuma xanthorrhiza) dan Sambiloto (Andrographis paniculata) merupakan tanaman obatIndonesia yang khasiatnya sudah banyak diketahui dan dimanfaatkan untuk kesehatan secara turun temurun,salah satunya sebagai imunostimulan atau peningkat daya tahan tubuh. Tujuan penelitian adalah untukmengetahui efikasi dari pengembangan formula herbal temulawak dan sambiloto dalam meningkatkan dayatahan tubuh, yaitu melalui peningkatan jumlah sel Th CD4+. Penelitian dilakukan menggunakan hewan cobamencit jantan galur ddY (deutch democratic Yokohama) yang diinduksi dengan 0,1 ml vaksin BCG i.p. padahari pertama dan ketujuh, pemberian ekstrak selama 10 hari, kemudian pada hari terakhir (H-10) dilakukanpengukuran kadar CD4+ menggunakan flowsitometer. Hewan coba dikelompokkan menjadi 7 kelompok,masing-masing kelompok terdiri dari 5 ekor. Kelompok pertama dan kedua diberi ekstrak sambiloto,temulawak secara oral dengan dosis 3 mg/20 g bb. Kelompok ketiga dan keempat formula ekstrak temulawakdan sambiloto dengan dosis 6 dan 30 g/20 gbb. Sedangkan kelompok lima, enam dan tujuh adalah kelompokkontrol positif, negatif dan pembawa. Hasil menunjukkan bahwa pemberian ekstrak maupun formulatemulawak dan sambiloto mampu meningkatkan jumlah sel Th CD4+ dibanding terhadap kontrol pembawa(p<0,05). Populasi sel Th CD4+ ekstrak sambiloto (4,19x103/ml) tertinggi dibandingkan dengan kelompoklainnya (II. 2,34x103/ml; III. 2,13x103/ml; IV. 2,70x103/ml), positif (2,56x103/ml), dan pembawa (3,21x103/ml). Dengan demikian dapat disimpulkan bahwa pemberian ekstrak maupun formula dari temulawak dansambiloto mampu meningkatkan jumlah sel Th CD4+ jika dibandingkan dengan kontrol pembawa (p<0,05).Kata kunci: temulawak, sambiloto, imunostimulan, sel Th CD4+
