Abstract


 

 

ABSTRACTSecang (Caesalpinia sappan) has been screen based on its activities showed posses anti-acne properties. Theactivities determination was done on anti-bacterial activity especially on Propionibacterium acnes, and P. acneslipase inhibitor. It has been reported that both brazilin and protosappain A were have antibacterial activities. Todetermine the capacity of secang as anti-inflamation agent, the TNF-α test has been conducted to secang extractusing THP-1 cell and also to the pure chemical contents that isolated from its extract. The result show that bothextract and pure chemical contents of secang were safe to THP-1 cell. Methanolic extract of 1 μg g/mL inhibitTNF- α on 31,6%, while brazilin, protosappanin A and sappanone N on the same concentration were be able toinhibit TNF- α on the following of 24.7%; 36.2% and 30.5% as well.Key words: Caesalpinia sappan, TNF-α, brazilin, protosappanin A, sappanone B

 

merupakantanaman yang memiliki potensi sebagai antijerawat berdasarkan aktivitasnya sebagai antibakteri terhadapPropionibacterium acnes, inhibitor P. acnes lipase dan antioksidan. Kami pun telah melaporkan bahwabrazilin dan protosappanin A merupakan senyawa yang memiliki ketiga aktivitas tersebut. Untuk mengetahuikemampuan secang sebagai antiinflamasi, uji TNF-α dilakukan terhadap ekstrak secang dan senyawa murniyang telah diisolasi dari secang. Uji TNF-α dilakukan menggunakan sel THP-1. Hasil penelitian menunjukkanbahwa baik ekstrak maupun senyawa murni dari secang tidak toksik terhadap sel THP-1. Ekstrak metanolsecang pada konsentrasi 1 μg/mL menghambat produksi TNF-α sebesar 31.6%, sementara brazilin, protosappaninA, dan sappanone B pada konsentrasi yang sama menghambat produksi TNF-α berturut-turut sebesar24,7%, 36,2%, dan 30,5%.
