POTENSI CAMPURAN MINYAK ATSIRI RIMPANG TEMULAWAK DAN DAUN CENGKEH SEBAGAI INHIBITOR PLAK GIGI

Abstract


Temulawak rhizome (Curcuma xanthorrlza) and cengkeh leaf CSyzygium aromaticum) essential oils have been reported to be potential inhibitors of planktonik growth and biofilm of S. mutans. This research aimed to reveal the potency of the essentialoils blend and the optimum compostition based on i15 inhibitory effect against planktonik growth and biofilm of S. mutans by an in vitro method. Blend composition of temulawak and cengkeh essential oils tested were 1:0; 0.75:0.25; 0.5:0. 1:0; 0.75:0.25; 0.5:0.5; 0.25:0.75 v/v; 0.25:0.75 v/v, respectively, in total concentration of 0.002%. The planktonic growth inhibitory assay was perfonned against S. mutans by micro dilution technique on nutrient broth media. Biofilm formation inhibition and degradation assay was condueted on BH!. Biofilm formation was indirectly assessed by staining with 1 % crystal violet. Data was obtained from plate reader in wavelength of 595 nm. The above assays were perfonned in triplicate. The results were used to detennine the SLD formula.
Results showed that the essential oils blend did not significantly influence the planktonic growth inhibition but significantly increase the biofilm formation inhibitory percentage as well as degradation. Optimum composition was detennined as temulawak essential oils 55 and cengkeh 45.

Abstrak

Minyak atsiri rimpang temulawak (Curcuma xanthorriza) dan daun cengkeh (Syzygium aromaticum) merupakan
inhibitor yang potensial dalam menghambat pertumbuhan planktonik dan biofilm karena bakteri S. mutans. Penelitian ini bertujuan untuk menggali potensi serta menentukan komposisi campuran minyak atsiri keduanya berdasarkan efek penghambatan pertumbuhan planktonik dan biofilm karena bakteri S. mutans secara in vitro. Campuran komposisi minyak atsiri yang diuji sebagai berikut: 1:0, 0.75:0.25, 0.5:0.1, 0.5:0.5, 0.25:0.75 v/v, dengan total konsentrasi 0.002%. Uji penghambatan pertumbuhan planktonik bakteri S. mutans
dilakukan dengan teknik mikrodilusi pada media nutrient broth. Penghambatan formasi biofilm dan uji degradasi dilakukan pada BHI. Formasi biofilm diuji secara tidak langsung dengan pengecatan 1% kristal violet. Data diperoleh menggunakan microplate reader panjang gelmobang 595 nm. Seluruh uji dilakukan dengan 3 kali pengulangan. Data digunakan untuk menentukan formula SLD. Hasil penelitian menunjukkan bahwa
campuran minyak atsiri tidak berpengaruh terhadap pertumbuhan planktonik secara signifikan, tetapi secara
signifikan dapat meningkatkan persentase penghambatan formasi dan degradasi biofilm. Komposisi optimum minyak atsiri adalah 55 temulawak dan 45 cengkeh.
