ANTICANCER ACTIVITY OF ZINGIBER OFFICINALE AND PIPER RETROFRACTUM EXTRACT COMBINATION ON HELA CELL LINE

Abstract


Penggunaan kemoterapi pada kanker menyebabkan efek samping karena berpengaruh dalam sintesis asam nukleat dan protein baik pada sel kanker maupun sel normal. Kombinasi tumbuhan dilakukan untuk meningkatkan aktivitas antikanker dan mengurangi efek samping kemoterapi. Rimpang jahe merah (Zingiber officinale cv Rubrum) dan buah cabe jawa (Piper retrofractum) secara empiris digunakan untuk mengobati kanker. Penelitian ini bertujuan untuk mengetahui aktivitas sitotoksik dan apoptotik kombinasi ekstrak jahe merah dan cabe jawa serta efeknya pada ekspresi p53 dan caspase 9 terhadap sel karsinoma servik HeLa. Aktivitas sitotoksik diuji dengan uji MTT [3-(4,5-dimetiltiazol-2-il)2,5-difeniltetrazolium], uji apoptotik dengan metode imunofluoresensi menggunakan fluorokrom etidium bromida dan akridin orange. Ekspresi p53 dan caspase 9 diuji dengan metode imunositokimia. Cisplatin digunakan sebagai kontrol positif. Hasil penelitian menunjukkan kombinasi ekstrak memiliki aktivitas terhadap sel HeLa dengan IC50 33.81 μg/ml, melalui mekanisme apoptosis, meningkatkan ekspresi gen supresor tumor p53 dan caspase 9.

Abstract

The use of chemotherapy for cancer cause side effects since its affect to synthesis of nucleic acids and proteins both cancer cells and normal cells. The combination of plants used to enhance the anticancer activity and to min:imize side effects caused by anticancer drug. Red ginger rhizome (Zingtberofflcinale ev. Rubrum) and java pepper fruit (Piper retrofractum) are empirically used as an anti cancer.
This study was conducted to investigate the cytotoxic and apoptotic activity of Zingiber officinale and Piper retrofractum extract and its effeet to p53 and caspase 9 expression on Hela cervical carcinoma cell. Cytotoxic assay was performed using MTT assay, apoptotic assay was performed by immunofluororescence method, using fluorochromes ethidium bromide and acridine orange. Expression of p53 and caspase 9 was examined by immunohistochemistry method. Cisplatin was used as positive control.
The result showed that extract combination has cytotoxic activity on Hela cell line, with IC50 - 33.81 ug/ml, through apoptotic mechanism, increase expression of p53 tumor suppressor gene and caspase 9.
